To investigate the effect of three red wines (RWs) from different growing areas and made from different grapes on asymmetric dimethylarginine (ADMA), an endogenous inhibitor of nitric oxide synthase, in young and senescent human endothelial cells (ECs). All RWs decreased ADMA levels, but 2-fold concentration of German RW was necessary to reach the same effect on ADMA compared to Italian RW and French RW without affecting the cell viability and morphology. The ADMA-lowering effect of RW was increased in senescent compared to young cells, accompanied by enhanced activity of the metabolizing enzyme: dimethylarginine dimethylaminohydrolase (DDAH) II, whereas the same amount in the upregulated protein expression of DDAH II and the downregulated protein expression of the synthesizing enzyme: protein arginine methyltransferase 1 was revealed. These effects were associated with decreased 8-iso-prostaglandin F and peroxynitrite formation, enhanced protein expression of NAD+-dependent class III histone deacetylase sirtuin (SIRT) 1, and downregulated protein expression of histone senescence factor p53. Blockade of SIRT1 activity abolished the effect of red wine on ADMA. These data are the first demonstration that RW by activating SIRT1 impairs synthesis and increases metabolism of ADMA. This effect of RW is accentuated in senescent cells probably due to enhanced DDAH activity.  相似文献

A neutral fraction (PS-SI) (0.3 g/L) with MW of 74 kDa, which contained galactose, arabinose, mannose, and glucose in the molar ratio of 1.0:0.6:0.4:0.2 was obtained by treatment of the whole polysaccharide extracted from red wine with cetrimide, followed by gel permeation chromatography. Spectroscopic and methylation analyses indicated that PS-SI is a mixture of neutral polysaccharides, consisting mainly of β (1→3)-linked galactopyranosyl residues, with side chains of galactopyranosyl residues at positions O-6. Arabinofuranosyl residues linked α (1→5), α-mannopyranosyl and glucosyl residues appear to be components of different polysaccharides. The in vitro antioxidant capacity of fractions of wine polysaccharide was studied by hydroxyl radical scavenging and ORAC assays. Fraction PS-SI presented the strongest effect on hydroxyl radicals (IC50 = 0.21).  相似文献

Lanthanide complexes are of great importance for their prospective applications in wide range of science and technology. Chiral lanthanide complexes can constitute stereo-discriminating probes in biological media, owing to the luminescent properties of the rare-earth ions. Sensitized emission with narrow bandwidth, having fast radiation rate and high emission quantum efficiency are the main perspective for synthesizing the complexes. Attention has been given on remarkable chirality with high dissymmetry factors (g = Δεext/εmax) of the complexes. For this purpose, beta-diketonato ligands with chiral BINAPO (1,1′-binapthyl phosphine oxide) ligand were chosen to achieve the goal. The complexes [Ln(TFN)3(S-BINAPO)](TFN = 4,4,4-trifluoro-1(2-napthyl)-1,3-butanedione), [Ln(HFT)3(S-BINAPO)] (HFT = 4,4,5,5,6,6,6-heptafluoro-1-(2-thienyl)-1,3-hexanedione) and [Ln(HFA)3(S-BINAPO)](hfa = hexafluoroacetylacetonate) (where Ln = Yb, Eu) were synthesized. The complex, [Eu(TFN)3(S-BINAPO)] gives strong red emission at 615 nm with narrow emission band (<10 nm) when excited by 465 nm light with quantum efficiency 86%. The dissymmetry factors (g = Δεext/εmax) corresponding to the 7F1 → 5D0 transition at 590 nm is 0.091 for [Eu(TFN)3(S-BINAPO)] and for [Yb(hfa)3(S-BINAPO)](hfa = hexafluoroacetylacetonate) corresponding to the 2F7/2 → 2F5/2 transitions is 0.12, are among the largest values for both Eu and Yb complexes to date, respectively. The Eu complexes, [Eu(HFT)3(S-BINAPO)] and [Eu(TFN)3(S-BINAPO)] are found to be spontaneously emissive, showing bright red emission, when placed in sunlight or even in the laboratory when light is switched on.  相似文献

Parrotfishes (family Scaridae) are important agents in marine bioerosion. Here, the feeding ecology of seven species of parrotfishes was studied on Egyptian Red Sea reefs. The most abundant species on both the reef flat and slope was Chlorurus sordidus. In contrast, C. gibbus had the lowest abundance on the reef flat, and Cetoscarus bicolor was the least abundant species on the reef slope. Scarus niger exhibited the highest feeding rate (98.9 bites 5 min− 1), followed by C. sordidus (76.5 bites 5 min− 1), whereas the rates for C. bicolor and C. gibbus were low (29.4 and 31.9 bites 5 min− 1, respectively). The daily feeding patterns of all seven species showed agreement in that activity was relatively constant over the day, with highest values in the early afternoon (1400 h) and a steady decrease until 1800 h. C. sordidus was more similar to S. niger and S. ghobban in showing somewhat higher activities in the morning (0800 h) followed by a slight decrease until noon. The average bite volumes of C. gibbus and C. bicolor were high (0.114 and 0.110 cm3, respectively), whereas S. niger had the lowest average value (0.002 cm3). Based on their feeding intensity, C. gibbus, S. ghobban and C. bicolor have high bioerosion rates on the Egyptian Red Sea reefs. Overall, S. ghobban is the most important bioeroder because it is more abundant than the other two species. All parrotfish species fed on dead coral and hard substrates which are rich in algae, but C. gibbus, C. bicolor and S. ghobban also fed on live coral on both reef zones; C. sordidus avoided live coral. The fresh scars on live coral were bigger than on dead coral because the three large parrotfish (C. gibbus, C. bicolor and S. ghobban) fed mainly on live corals.  相似文献

We previously demonstrated that canine erythrocytes express the P2X7 receptor, and that the function and expression of this receptor is greatly increased compared with human erythrocytes. Using 86Rb+ (K+) and organic cation flux measurements, we further compared P2X7 in erythrocytes and mononuclear leukocytes from these species. Concentration response curves of BzATP- and ATP-induced 86Rb+ efflux demonstrated that canine P2X7 was less sensitive to inhibition by extracellular Na+ ions compared to human P2X7. In contrast, canine and human P2X7 showed a similar sensitivity to the P2X7 antagonists KN-62 and Mg2+. KN-62 and Mg2+ also inhibited ATP-induced choline+ uptake into canine and human erythrocytes. BzATP and ATP but not ADP or NAD induced ethidium+ uptake into canine monocytes, T- and B-cells. ATP-induced ethidium+ uptake was twofold greater in canine T-cells compared to canine B-cells and monocytes. KN-62 inhibited the ATP-induced ethidium+ uptake in each cell type. P2X7-mediated uptake of organic cations was 40- and fivefold greater in canine erythrocytes and lymphocytes (T- and B-cells), respectively, compared to equivalent human cell types. In contrast, P2X7 function was threefold lower in canine monocytes compared to human monocytes. Thus, P2X7 activation can induce the uptake of organic cations into canine erythrocytes and mononuclear leukocytes, but the relative levels of P2X7 function differ to that of equivalent human cell types.  相似文献

摘要 目的:探讨祛毒汤联合红光照射治疗促进湿热下注型低位单纯性肛瘘患者术后创面愈合的临床疗效。方法:采用随机数字表法将西安市中医医院肛肠科2019年12月至2022年12月期间收治的116例湿热下注型低位单纯性肛瘘患者分为观察组(58例,红光照射、祛毒汤熏洗、常规治疗)和对照组(58例,红光照射、常规治疗)。对比两组临床疗效、创面愈合情况、血清炎症因子、血清生长因子变化情况。结果:与对照组相比,观察组创面愈合率更高,创面愈合时间更短,临床总有效率更高(P<0.05)。与对照组治疗后相比,观察组同时间点视觉模拟评分法(VAS)评分、创面渗出评分、创面肉芽生长情况评分更低,超敏C反应蛋白(hs-CRP)、降钙素原(PCT)、白细胞介素-6(IL-6)更低,碱性成纤维细胞生长因子(bFGF)、表皮生长因子(EGF)、转化生长因子β(TGF-β)更高(P<0.05)。结论:祛毒汤联合红光照射治疗可促进湿热下注型低位单纯性肛瘘患者术后创面愈合,可调节炎症因子和生长因子水平。  相似文献

目的:构建弗氏2a志贺氏菌2457T株yciD基因缺失突变体,以研究yciD基因的功能。方法:根据弗氏2a志贺氏菌2457T株基因组全序列,采用Red重组系统对yciD基因进行缺失,并经PCR和SDS-PAGE证实;对野生株和突变株的生长状态及生化反应进行比较研究。结果:构建了弗氏2a志贺氏菌2457T株的yciD基因缺失突变株2457TΔyciD,该突变株外膜蛋白样品中缺失了一条相对分子质量与从yciD基因推导的蛋白相当(约22000)的蛋白带。该突变株比野生株生长快,利用葡萄糖和甘露醇的能力也比野生株大为增强。结论:获得了弗氏2a志贺氏菌2457T株的yciD基因缺失突变株。  相似文献
